Sādhana means the activity by which we can attain, we can achieve that goal of life. This is called sādhya-sādhana.
Lecture on BG 1.43 -- London, July 30, 1973:

Pradyumna (leads chanting, etc.):

utsanna-kula-dharmāṇāṁ
manuṣyāṇāṁ janārdana
narake niyataṁ vāso
bhavatīty anuśuśruma
(BG 1.43)

Translation: "O Kṛṣṇa, maintainer of the people, I have heard by disciplic succession that those who destroy family traditions dwell always in hell."

Prabhupāda: So family tradition, whose family tradition? Manuṣyāṇāṁ. Family... Manuṣyāṇāṁ, those who are human being, they have got family and family rituals, kula-dharma, jāti-dharma. Just like cātur-varṇyaṁ mayā sṛṣṭaṁ guṇa-karma-vibhāgaśaḥ (BG 4.13). This is meant for the human being, manuṣyāṇāṁ, not for the animals. So unless the human society accepts this principle of varṇāśrama-dharma ordained by Kṛṣṇa, the Supreme Lord, he is not considered amongst the human being. They are as good as animals because there is no regular, systematic principles of how to live, a human being. Therefore in the Viṣṇu Purāṇa... When Caitanya Mahāprabhu was talking with Rāmānanda Raya, so Caitanya Mahāprabhu inquired, "What is the actual aim of life?" Sādhya-sādhana (CC Madhya 8.118). Sādhya and sādhana. "What is the aim?" Sādhya means what for this human form of life we have got. That is called sādhya, the goal, the goal of life. And sādhana. Sādhana means the activity by which we can attain, we can achieve that goal of life. This is called sādhya-sādhana. The Darwin theory, although they are giving some idea of progressive evolution, but he does not know what is the goal of life, why this progress is there. That these rascals they do not know. Simply just imagining, taking some whims from the Padma Purāṇa, Brahmā Vaivarta Purāṇa. Aṣitiṁś catvarāṁś caiva, jīva-jātiṣu, lakṣāṁs tān jīva-jātiṣu. There are 8,400,000 types of jīva-jātis. Aṣitiṁś catvarāṁś caiva lakṣāṁs tān jīva-jātiṣu. So this is already there. Brahmādbhiḥ. In this way the living entity is wandering in the cycle of transmigration of the soul. So from this, Mr. Darwin might have taken some hints, but he could not explain properly. He could not catch up that brahmādbhiḥ, who is wandering. So it is a chaos.

So anyway, why this living entity is wandering, not fixed up? So there must be some goal. He is hankering after that. There must be some goal of life. To achieve that goal of life that is called sādhya. Why we are struggling here for happiness or something else? We are struggling. So this question was raised by Caitanya Mahāprabhu, that "What is the goal of life?" Unless there is goal of life, why there is struggle? Why... There must be some goal of life, sādhya. And sādhana. Sādhana means the means by which we can achieve that goal of life. That is called sādhana, sādhana. So Rāmānanda Rāya quoted... Because when there is talk between two learned persons, they... Just like nowadays it has become a fashion: "In my opinion," "I think in this way." What, nonsense, what you can think? What is your knowledge? But he is very proud. Everyone like that. The other day, that Sir Alistair? Alistair Hardy came. He also said, "It is my opinion." Nobody thinks that he is a nonsense number one; what is the value of his opinion? Nobody thinks. But this is the Vedic principle. Even Caitanya Mahāprabhu, He, when He was asked by Prakāśānanda Sarasvatī that "You are a sannyāsī. You do not engage Yourself in the study of Vedānta. You are simply chanting Hare Kṛṣṇa, Hare Kṛṣṇa." So... Because a sannyāsī is supposed to be always reading Vedānta philosophy especially and all other philosophy.

Sādhana means cultivating something with a aim of objective.
Lecture on SB 1.2.28-29 -- Vrndavana, November 8, 1972:

When Rāmānanda Rāya was questioned by Śrī Caitanya Mahāprabhu: "What is sādhya and sādhana, what is human duty, sādhya, and what is the end of sādhana...?" Sādhana, sādhana means cultivating something with a aim of objective. So this question was put to Rāmānanda Rāya by Śrī Caitanya Mahāprabhu: "What is the aim of human life and what is the process of executing that will?" So immediately Rāmānanda Rāya said that varṇāśramācāravatā. The beginning of human life is to execute the varṇāśrama-dharma. Varṇāśramācāravatā puruṣeṇa paraḥ pumān (CC Madhya 8.58). Why varṇāśrama? Now viṣṇur ārādhyate. If you properly execute your varṇāśrama-dharma... Actually, it is now pervertedly spoken as Hindu dharma. Hindu dharma is a vague term. Real dharma is varṇāśrama-dharma. Hindu dharma we don't find, any Vedic literature. Neither in the Bhagavad-gītā. It is a, a nomenclature given by the Muhammadans—"Hindus." From Sindhu, "Hindu." Anyway, now we are known as Hindus. The "Hindu" is a vague term. Real term is varṇāśrama, varṇāśrama, four varṇas and four āśramas. This is dharma and this is given by God Himself. Just like Kṛṣṇa says, cātur-varṇyaṁ mayā sṛṣṭaṁ guṇa-karma-vibhāgaśaḥ (BG 4.13). It is given by God. You cannot manufacture dharma, religion. No. Dharmaṁ tu sākṣād bhagavat-praṇītam: (SB 6.3.19) "Dharma means the codes, the regulations given by God." That is dharma. That is dharma. Otherwise, it is not dharma.

Sādhya means the objective thing, and sādhana means the process.
Lecture on SB 5.6.4 -- Vrndavana, November 26, 1976:

The Vedic culture is actual civilization, varṇāśrama-dharma. That is the beginning of civilization. Without Vedic culture and varṇāśrama-dharma it is animal civilization. When Rāmānanda Rāya and Caitanya Mahāprabhu was talking, Caitanya Mahāprabhu inquired from Rāmānanda Rāya, "What is the beginning of life?" Sādhya-sādhana."What is the object, objective of the ultimate goal of life, and how that objective of life is attained?" Sādhya-sādhyana. Sādhya means the objective thing, and sādhana means the process. Sādhya-sādhana. So Rāmānanda Rāya, he said, varṇāśrama. Varṇāśrama. That is the beginning of life. Because varṇāśrama means... The life is... Human life is meant for understanding the Supreme Lord. That is human life. The cats' and dogs' life is not meant for understanding God; it is not possible either. They cannot understand.

For a sādhana, (?) means a neophyte devotee, he is advised to give up these low grade habits—kāma, krodha, lobha, moha, mada, mātsarya.
Lecture on SB 7.9.1 -- Mayapur, February 10, 1977:

Pradyumna: Śrīla Narottama dāsa Ṭhākura has sung in his prema-bhakti-candrikā, 'krodha' bhakta-dveṣi jane: anger should be used to punish a demon who is envious of devotees. Kāma, krodha, lobha, moha, mada and mātsarya—lust, anger, greed, illusion, pride and envy—all have their proper use for the Supreme Personality of Godhead and His devotee. A devotee of the Lord cannot tolerate blasphemy of the Lord or His other devotees, and the Lord also cannot tolerate blasphemy of a devotee. Thus Lord Nṛsiṁhadeva was so very angry that the great demigods like Lord Brahmā and Lord Śiva and even the goddess of fortune, who is the Lord's constant companion, could not pacify Him, even after offering prayers of glorification and praise. No one was able to pacify the Lord in His anger, but because the Lord was willing to exhibit His affection for Prahlāda Mahārāja, all the demigods and the others present before the Lord pushed Prahlāda Mahārāja forward to pacify Him.

Prabhupāda: So, in the material world, kāma, krodha, lobha, moha, mada, and mātsarya, these things are condemned. And for a sādhana, (?) means a neophyte devotee, he is advised to give up these low grade habits—kāma, krodha, lobha, moha, mada, mātsarya. But people may question, "Wherefrom these lowgrade qualities came? Kāma is low grade, admitted, but wherefrom it came?" In the Vedānta-sūtra we get the reply, janmādy asya yataḥ: (SB 1.1.1) "Everything that we experience, it comes from God." So... (aside:) You stand that side. You are standing. They can stand.

"In the transcendental realm of Vraja (Vraja-dhāma) one should serve the Supreme Lord, Śrī Kṛṣṇa, with a feeling similar to that of His associates, and one should place himself under the direct guidance of a particular associate of Kṛṣṇa and should follow in his footsteps. This method is applicable both in the stage of sādhana (spiritual practices executed while in the stage of bondage) and in the stage of sādhya (God realization), when one is a siddha-puruṣa, or a spiritually perfect soul."

Devotee (1): Sādhana means accepting discipline.

Prabhupāda: Eh, yes. Do you think that solving the problem of birth, death and old age and disease is very easy task? (chuckles) You have to very, very strongly... Those who do not care, that is another thing. They are under these laws, going on. Life after life, they are changing body, again birth, again death, again in the womb of mother. They do not take it seriously, how risky life is that. They do not take it. "Oh, what is that?" Now, in Kali-yuga, the mother is killing the child. Even within the womb of mother is not safe. This is going on. So they do not take that how this way of life is risky and uncomfortable. They do not take it seriously. To enter again into the womb of mother, how difficult job and troublesome it is, they do not understand it. For want of education.
